Could someone who has played Destruction Derby via serial linkup tell
me if both computer need to have the CD. I'm taking my computer to a
friend's house this weekend and I'm curious if we'll be able to play
this with only 1 copy of the game.

You need the another copy. BTW you if your mate is going to buy a copy
you should both be aware that you need a 16550 UART on your I/O card,
anything less (8450 usually) it won't even try to link.

IMHO it's not worth buying DD for a linkup. Me and a mate tried it and
was very dissappointing. The Bowl is flawed on a 2 player (takes ages to
get anywhere) and for some reason with any event if we had other cars on
the track the game would crash after a few seconds. There's probably a
patch for this somewhere.

Play Descent or C&C instead.
