Right-click on "My Computer", select Properties.
Select the "Device Manager" tab, and choose "View devices by connection".
Open a few things up in the tree until you find "PCI Bus". Open this up
and look for something mentioning "AGP Controller". eg mine says "Intel blah
blah blah Processor to AGP Controller". If it's there, you have an AGP slot.

I would have thought that the best way is to go into the BIOS settings
when the machine starts up, normally by pressing the DEL key or
something similar (it should say which key to press to run SETUP when
the machine boots up).  From in the BIOS settings, if your board has an
AGP slot then there should be an option to enable it in the BIOS.  If
there is absolutely no reference to AGP anywhere then you don't have
AGP.

PS.  I know this is rather a late follow up, but my ISP went***up
with its News server for 4 days.
